چکیده

OH OH Two main classes of aldolases are found in Nature. Type I aldolases activate the nucleophilic partner by forming an imine as an intermediate; this type of enzyme is found in animals and higher plants. Type II aldolases occur in microorganisms and they require R 2 to be a hydroxy group. These enzymes feature a Zn 2+ ion that promotes the reaction by forming a chelate with both the carbonyl and an α-hydroxy group on the nucleophilic partner. Aldolases are highly stereoselective; the particular absolute and relative stereochemistry shown below is just for illustration. Aldolases perform many important metabolic reactions, in both the forward and reverse directions. OH fructose bisphosphate aldolase β-D-fructose-1,6-phosphate D-glyceraldehyde 3-phosphate dihydroxyacetone phosphate NH 2 glycine acetaldehyde pyruvate phosphoenolpyruvate dihydroxyacetone(phosphate) Aldolases will accept a wide variety of aldehydes as electrophiles, but the scope of the nucleophilic partner is usually limited to the natural substrate or a very close analog. * denotes stereocenter formed An engineered aldolase is being used on an industrial scale to produce the side chain of the statin drugs, widely prescribed for reducing the risk of cardiovascular issues.

Enhanced aldolase activity and glycolysis as short term effects of hydrogen cyanide for the release of dormancy in walnut kernels

Seed dormancy removal by cold stratification is accompanied by the development of gluconeogenic competence. Although hydrogen cyanide can stimulate the germination of many herbaceous dormant seeds and increase gluconeogenesis in long term, its short-term effects on sugar metabolism require further investigation. Accordingly, an experiment in the form of complete randomized design was carried ou...

Levels of C-reactive protein, creatine kinase-muscle and aldolase A are suitable biomarkers to detect the risk factors for osteoarthritic disorders: A novel diagnostic protocol

Background: C-reactive protein (CRP), creatine kinase-muscle (CK-MM) and aldolase A (AldoA) levels are predicted to be realistic biomarkers of osteoarthritic disorders (OADs). The objective of the study was to evaluate the levels of CRP, CK-MM, and AldoA and determine their correlations with risk factors such as inflammation, muscle degeneration, and skeletal muscle damage for OADs. Methods: B...

Action of cathepsin D on fructose-1,6-bisphosphate aldolase.

Cathepsin D inactivated aldolase at pH values between 4.2 and 5.2; the chloride, sulphate or iodide, but not citrate or acetate, salts of sodium or potassium accelerated the rate of inactivation. Cathepsin D cleaved numerous peptide bonds in the C-terminus of aldolase, but the major site of cleavage in this region was Leu354-Phe355. The most prominent peptide products of hydrolysis were Phe-Ile...

Automated procedure for serum aldolase estimation. Fructose-1, 6-diphosphatase and fructose-1-phosphatase.

SERUM ALDOLASE ESTIMATION 1S one of the principal diagnostic tests used in the elucidation of muscle disease (2-6). In myocardial infarction (7), liver disease (8), and carcinomatosis (9), the serum aldolase estimation is a useful ancillary aid to diagnosis. The usefulness of the test has been enhanced since the differentiation 1)etween muscle and liver aldolase by use of two substrates-fructos...

Detection of aldolase activity on polyacrylamide gels: application to 2-keto-4-hydroxyglutarate aldolase.

A method is described for the detection of 2-keto-4-hydroxyglutarate aldolase activity after electrophoresis of the enzyme on polyacrylamide gels. When gels are incubated with substrate (2-keto-4-hydroxyglutarate), activity is seen as a yellow-colored band due to interaction’ of the product (glyoxylate) with ortho-aminobenzaldehyde and glycine. Positive results have been obtained using either c...

Targeting of several glycolytic enzymes using RNA interference reveals aldolase affects cancer cell proliferation through a non-glycolytic mechanism.

In cancer, glucose uptake and glycolysis are increased regardless of the oxygen concentration in the cell, a phenomenon known as the Warburg effect. Several (but not all) glycolytic enzymes have been investigated as potential therapeutic targets for cancer treatment using RNAi. Here, four previously untargeted glycolytic enzymes, aldolase A, glyceraldehyde 3-phosphate dehydrogenase, triose phos...
